Phase I Drug Metabolism

It includes oxidation (via cytochrome P450), reduction, and hydrolysis reactions.phase I reactions convert a parent drug to more polar (water soluble) active metabolites by unmasking or inserting a polar functional group (-OH, -SH, -NH2).

Drug Toxicology

Drug Toxicology is defined as the scientific study of the adverse effects of drugs on the living organism. It involves observing and reporting symptoms, mechanisms, detection and treatments of drugs, in particular relation to the poisoning of humans. It includes drugs that are synthesized for medical use by humans. These substances may produce toxic effects in living organisms including disturbance in growth patterns, discomfort, disease and death.

Biochemical Metabolism

Biochemical Metabolism is a pathway having series of chemical reactions occurring within a cell. In a pathway, the initial chemical (metabolite) is modified by a sequence of chemical reactions. Cytochrome 450 Drug Metabolism

Cytochromes P450 (CYPs) belong to the superfamily of proteins containing a heme cofactor and, therefore, are hemoproteins. CYPs use a variety of small and large molecules as substrates in enzymatic reactions. CYPs are the major enzymes involved in drug metabolism,most drugs undergo deactivation by CYPs, either directly or by facilitated excretion from the body. Also, many substances are bioactivated by CYPs to form their active compounds.

Liver Drug Metabolism

The liver is the principal site of drug metabolism. Although metabolism typically inactivates drugs, some drug metabolites are pharmacologically active sometimes even more so than the parent compound. An inactive or weakly active substance that has an active metabolite is called a prodrug, especially if designed to deliver the active moiety more effectively.

Drugs metabolised by CYP2D6

CYP2D6 gene encoded in humans for the enzyme Cytochrome P450 2D6.It is expressed in the liver and in areas of CNS. It is responsible for the metabolism and elimination of approximately 25% of clinically used drugs, in a process referred to as O-demethylation. This enzyme also metabolizes several endogenous substances such as hydroxytryptamines and neurosteroids.
