Biography :

Irina Grishina obtained her Ph.D in Biochemistry at Colorado State University based on experimental and theoretical studies in protein spectroscopy and thermodynamics. She continued on to postdoctoral training in molecular genetics of cell cycle with Roger Brent at Harvard Medical School and James Maller at University of Colorado Health Sciences Center. Since 2001, Grishina engaged in studies in organogenesis at New York University, and established a program on urogenital development in mouse models at the Department of Urology. Grishina’s laboratory is interested in defining the roles of developmental signaling factors which, in collaboration with steroid hormones, regulate growth, differentiation and tissue homeostasis of the prostate, urethra and external genitalia. Grishina’s studies are directed to elucidate the etiology of urogenital diseases and malformations, in particular, benign prostate hyperplasia, rectourethral fistulas and hypospadias.

 Biography
Research Interest :

1. Development and disease of the prostate gland. We have shown that developmental ligand of transforming growth factor family, Bone morphogenetic protein 7 (Bmp7), inhibits prostate branching and restricts cells positive for Notch signaling to prostate buds (Grishina et al., http://www.ncbi.nlm.nih.gov/pubmed/16324690). Gain of Notch signaling in the prostate results in abnormal proliferation of the basal and myoblast progenitor cells, which leads to benign prostate hyperplasia (BPH) by 3 months of mouse age (Wu et al., 2011a, http://dx.doi.org/10.1016/j.ydbio.2011.05.659). This disease is mediated by the Notch-dependent downregulation of the tumor-suppressor Pten, improved maintenance of p63-positive basal cells, and cell fate shift favoring myoblast over fibroblast lineages. Loss of Notch signaling during prostate development results in abnormal patterning of basal cells which acquire luminal characteristics similar as in prostate cancer. Our further studies are aimed to elucidate the role of Bmp and Notch signaling in benign prostate disease.
2. Development and malformations of the cloaca and genital appendage. Current models for cloacal septation in mammalian embryo are derived primarily from anatomical observations and many postulates require clarification and experimental proof. Our studies are aimed to clarify the genetic and cellular mechanisms of cloacal and genital development. We found that signaling by Bmp7 is essential for cloaca septation and for formation of the ventral part of the genital appendage (Wu et al., 2009, http://www.ncbi.nlm.nih.gov/pubmed/19159697). Our previous and on-going studies show that cloacal epithelium is the tissue that undergoes extensive remodeling during cloacal septation. We are further investigating the role of Bmp and Wnt signaling in during the early stages of cloacal and genital development.

After obtaining my degree in Biochemistry (July 2001; Oviedo University, Spain) I worked at the Centro de Investigaciones Biológicas (CSIC; Madrid, Spain) where I acquired basic knowledge on myeloid cell biology and routine techniques (Sep. 2001 - Dec 2002). Then I obtained a fellowship from the Spanish Ministry of Science and Technology to do my Doctoral Thesis at the Centro Nacional de Biotecnología (CSIC; Madrid, Spain) on a specific feature of myeloid cells, such as the pattern of expression and functionality of chemokine receptors (Jan. 2003 – Feb. 2007). Later, I was awarded by the Theleton Institute for gene therapy with a postdoctoral fellowship (March 2007 – Aug. 2008) to improve my knowledge in lentiviral platforms used on myeloid cells in Prof. Luigi Naldini´s lab (Ospedale San Raffaelle, Milan, Italy) who is considered one of the best specialists in the field. I continued with a postdoctoral fellowship from Alfonso Martin Escudero foundation to directly apply our new developed lentiviral vectors and protocols optimized for myeloid cells on the study and genetic modification of macrophages associated to pathologies, specially tumors, at Istituto Clinico Humanitas (Sep. 2008 – Sep. 2010; Milan, Italy). During this two years fellowship I identified the c-Myc transcription factor as one of the main controllers of the phenomenon of macrophage polarization. During my postdoc in Milan, I performed a short-stay in Dr. Rodrigo Mora’s lab at Massachusetts Harvard Medical School to study the possible expression of c-Myc in different myeloid cells such as gut-associated dendritic cells (granted by the European macrophage and dendritic cell society). In October 2010, I was awarded with a contract from the Spanish Ministry of Science and Innovation (Juan de la Cierva programme) to continue my work in the field of macrophages associated to cardiovascular diseases at the Centro National de Investigaciones Cardiovasculares (CNIC; Madrid, Spain).

Department of Pediatrics
Division of Paediatric Endocrinology
McMaster University

Canada

Biography :

I became interested in academic Paediatric Endocrinology when I was a medical student. This decision was partly influenced by conducting research under the supervision of Professor Hilary Hoey, Head of Department of Paediatrics in Trinity College Dublin, and a leading figure in Paediatric Endocrinology in Ireland.
My work with her focused on studying the effects of treatment of patients with Turner syndrome with Growth Hormone with and without the anabolic steroid Oxandrolone, and we showed that in our group there was modest benefit of this combination in maximizing final height. In this stimulating environment, I excelled in my study of Paediatrics during medical school and was awarded the first prize in the final Paediatric exam [the O’Donohue Medal].

Few years later, I had the privilege of working with her again at a more senior level in the department of Endocrinology, Diabetes and Growth in Trinity College Dublin; in addition to regular diabetes and endocrine clinics, I was involved in managing patients in multidisciplinary obesity clinics where we delivered comprehensive clinical care and evaluated the various risk factors and complications that these children had. As this clinic was staffed by an exercise specialist, social worker, dietician, psychologist and a psychiatrist, it gave me the opportunity to deal first hand with these complex patients. Since then, I developed my interest in studying childhood obesity and its co-morbidities.

Following graduation and internship, I started my Paediatric training. The training program in Ireland is composed of two phases: General Professional Training and Higher Specialist training; at the end of this program I was awarded the Certificate of Satisfactory Completion of Specialist Training in Paediatrics that allows me to practice as an independent specialist in Europe. During the latter part of Higher Specialist Training, I did one year of Paediatric Endocrinology at the two largest academic Hospitals in Dublin. In addition to Professor Hoey, I worked with Dr. Colm Costigan, a Canadian trained Paediatric Endocrinologist and an exceptional teacher, mentor and person.  He provided guidance that was essential for me to mature in my understanding of the complexity of the specialty. During my training, I also conducted two important studies; the first looked at outcomes in infants of diabetic mothers in the Coombe Women’s Hospital, a tertiary, 7500 births per annum Perinatal centre; we showed that due to improved maternal diabetes care, the outcomes in these infants were very similar to those born to mothers with no diabetes during pregnancy.
The second study looked at the neonatal hypothyroidism screening program in south of Ireland under the supervision of Dr. John McKiernan in University College Hospital in Cork, Ireland. We were able to show that the quality of care for these infants remained excellent over a 10-year period. This project allowed me to appreciate the importance of assessing the quality of service delivery.

I also recognized from an early stage the importance of being a good manager, delegator, communicator, mediator, leader, and collaborator. In order to strengthen my knowledge in the area of health care management, I enrolled in a one-year, part time diploma course that was offered by the Royal College of Surgeons of Ireland and the Institute of Public Administration, a body mandated with training the next generation of leaders in Irish healthcare system. This allowed me to improve my managerial, administrative and leadership skills.

During my clinical training, I found myself interested in understanding mechanistic details regarding disease causation. As a result, I approached my mentors at the time who offered me the opportunity to receive formal research training, and I chose to do this in one of the most prestigious research institutions in the world, St. Jude Children’s Research Hospital, Memphis, Tennessee.  I embarked on a three-year full time research fellowship, during which I received excellent training in molecular biology and developed a far deeper understanding of the scientific process; my work was focused on understanding mechanisms of gene expression through the study of animal and cell models. This was a tremendously exciting and busy time that allowed me to truly grow and develop not only as a scientist but as a person. Following completion of my fellowship in St. Jude, I moved to Canada to pursue further training in Paediatric Endocrinology and to continue to do research. Again, I chose one of the top clinical and research establishments in the world, The Hospital for Sick Children and the University of Toronto.

Biography :

In 1990 I graduated in Biological Science at the University of Valencia and remained there to undertake a PhD under the supervision of Consuelo Guerri at Research Institution (Instituto de Investigaciones Citológicas), which I completed in 1996. During my PhD my dissertation was “Changes of astroglia intermediate filaments gene expression during rat brain development: Effect of alcohol exposure”. I analyze the events which occur during the early stages of astrogliogenesis during brain development, using “in vivo” and “in vitro” experiments. I did experiments in cells in culture primary as radial glia (stem cells in brain), astrocytes and neurons.
In 1997 I joined Eva Qwanstrom’s group at the Hallamshire Hospital (University of Sheffiel) in Sheffield, UK and spent three years involved in the identification of an adhesion-regulated subunit of the interleukin-1 (IL-1) receptor complex. I was working in immunology, cytokines, inflammation processes and matrix regulation of IL-1 responses.
In 2000 I returned to Spain at Department of Physiology, Medicine Faculty of Valencia. University of Valencia and I were appointed to a part-time position as Lectureship. In 2004 I was appointed to a fixed-term position as permanent University Lecturer at this department in the University of Valencia. During this time I was involved in the mechanisms of oxidative stress in the generation of Alzheimer’s disease with Jose Viña’s group. Also at this time I developed my lectures with Luis Such’s group with who I learned a lot about physiology and how to do a correct lecturer to pupils in my our department.

At this moment I work in Alzheimer’s diseases and in inflammation and oxidative stress mechanisms. Also I would like to work in precursor cells and the subsequently ability to further differentiate into neurons and astroytes inside the brain. I order a grant from my sanity government and I still waiting for the resolution grant, about stem cells and brain.
