aThe limit-dilution calculation was based on a Poisson probability distribution: ln (y) = rx + ln (a), where y is the ratio of non–tumor-generating injections, x is the number of cells injected, and r is the frequency of CSC; a is the y intercept and it was assumed that with no cells injected, no tumors are formed (therefore at 100% non–tumor generation, a = 1). The frequency of CSCs (r) was calculated from the slope of line best fit to the data points [56,57].
