Figure 7: Influence of GM6001 and blebbistatin on degradation characteristics of MDA-MB-231 cells. MDA-MB-231 cells were cultured on 10 μg/cm2 collagen-coated surfaces for 24 hours in the presence of 5 μM GM6001 or 5 μM blebbistatin. While matrix degradation was assessed by staining untreated and drug-treated samples with collagen antibody, invadopodia formation was assessed by co-staining with F-actin and cortactin. (A) Representative images of collagen staining in untreated and drug-treated sanples. Scale bar=200 μm. (B) Matrix degradation was negligible in both GM6001-treated and blebbistatin-treated cells. Bars indicate statistical significance (*p < 0.001). (C) High magnification (40x) images of F-actin/cortactin staining in control and drug-treated cells. Scale bar=50 μm. (D) Number of invadopodia was significantly reduced in GM6001-treated and blebbistatin-treated cells. Bars indicate statistical significance (*p < 0.001). |
