Adiponectin (Acrp30) expression levels was measured by real time PCR in RNA isolated from adipose tissue from 14 week old, male, ob/ob mice following 4 weeks of treatment with compound 1. N=5 for each group. AMPK activity was measured by Western blot analysis of phosphorylated AMP levels which was normalized to AMPK total protein levels. Antibodies to phospho-AMPK-alpha, (Thr172), and AMPK-alpha were purchased from Cell Signaling, Inc. Antibodies (80 ug/ml) were diluted 1:1000. Percent change compared to average of vehicle control animals is shown for adiponectin expression and AMPK activity. Data is the average of 5 tissues per group, and represent means ± SD. P values were calculated using two sample, paired t test.
