Figure 8: Lipid-modified proteins have been studied using bioorthogonal reactions. After metabolic labeling with alkyne (in green) or azide-containing lipids (in purple) and cell lysis, proteins were labeled by means of Staudinger ligation or CuAAC with biotinylated or fluorescent tags for further analysis and identification. Alternatively, these methods can be also used prior to cell lysis for cell imaging in fixed cells. |
