Figure 2: Macrophages polarized to the M1 state with IFN-γ are better at hydrogen peroxide scavenging than IL-4 treated M2 macrophages. NR8383 cells were treated with IFN-γ as above. They were then treated with glucose oxidase (GOX) 5 mU/ml to produce a steady level of hydrogen peroxide (H2O2). After four hours, relative H2O2 scavenging was assessed by Amplex Red Assay by subtracting the MFI for the treated samples from a cell-free sample treated with GOX in parallel. Cells polarized to the M1 state showed a significant improvement H2O2 scavenging. Data presented as mean ± SEM, n=4. *p<0.05 compared with IL-4 treated cells. |
