Figure 6: Effect of phototherapy on cytokines. Supernatant from U937 cells cultured with LPS (1 μg.mL-1) and treated or not with phototherapy (4.5 J/cm2) were collected in order to analyze the mRNA and the protein concentration of TNF (A and B), IL-1β (C and D), IL-6 (E and F), IL-8 (G and H) (pro-inflammatory interleukins) and IL-10 (I and J) (anti-inflammatory interleukin). The cells received laser therapy one hour after the addition of LPS in the culture medium. One hour before LPS, U937 cells were pretreated with 20 mM Nacetylcysteine (NAC). Twelve hours after U937 cells stimulation with LPS the mRNA and the protein concentration of inflammatory mediators were determined by Enzyme Linked Immuno Sorbent Assay (ELISA) and RT-PCR, respectively. Each bar represents mean ± S.D. of three independent experiments. Statistical differences were considered when p<0.05. |
