Signaling is triggered by the interaction with Notch ligands, which induces proteolytic cleavage of the Notch and leads to the nuclear translocation of the Notch intracellular domain (ICN). The intracellular Notch displaces the co-repressors (CoR) from CSL and recruits co-activators (CoA) to activate the expression of target genes.
