Figure 1: PPARγ agonists attenuate estradiol-dependent growth of human breast cancer cells. A, Estradiol stimulation of breast cancer cell proliferation is attenuated by PPARγ. MCF-7, ZR-75 and T47D cells were seeded in 96-well plates and treated with vehicle (Veh) (ethanol), 17β-estradiol (E2) (10 nM), rosiglitazone (Rosi) (1 μM), or E2+Rosi for 92 h and then assayed for cell growth. Graphical data are represented as % increase in cell number over vehicle (set at 100%). Data points represent the average of quadruplicate samples for each condition in a representative experiment. Proliferation assays were optimized for each cell type as described previously. B, MDA MB-231 cells were seeded in 96-well plates and treated as above in the absence or presence of GW9662 (1 μM) for 92 h. Cell proliferation (quadruplicate samples per treatment) was assayed as described above.
