LASV NP fragments were inserted using the Acc65I and Sal I sites in the polylinker sequence (red box). The original YF17D translation initiation codon has been inactivated. Translation of the recombinant polyproteinstarts at the second AUG codon. The NH2-terminus of the YF17D C protein starts with Pro-Met-Val (PMV). Proteins encoding DNA fragments that are cloned in the correct ORF relative to the remainder of the YF17D polyprotein will be fused to the 3X Flag epitope (blue box) to allow easy identification. Brown box represents the so-called FMDV 2A cleavage site.
