Figure 3: Molecular symptoms of human recombinant DNases’ directed suicide of proliferating stem cells. Human autologous pluripotent induced stem cells (hapiSCs) were transfected with the SSEA-4 antibody guided vectors carrying coding sequences for DNASE1, DNASE1L3, DNASE2, and DFFB controlled by POLA promoter. Early signs of these transgenes’ expression, concurrent with G1/S transition, were manifested by externalization of phosphatidylserine (A) and membrane blebs corresponding to those shown in the figure 2. Zernicke’s phase contrast served as the reference (B). Transgenic expression of the human recombinant DNases resulted in complete collapse of chromatin architecture (C). |
