3-D Structure Determination

Biomolecules are very small to see in detail even by most cutting-edge light microscopes. The methods that the structural biologists use to determine their structures in general involve the measurements on huge numbers of identical molecules at the same time. Some of the best methods include X-ray crystallography, Cryo-Electron Microscopy and Nuclear Magnetic Resonance. Very often scientists use them to study the "native states" of biomolecules.

The main techniques used for 3 D structure determination of biomolecules are X-ray crystallography, Nuclear Magnetic Resonance and Cryo-electron microscopy. Other than these main techniques, a number of other techniques are used as well such as ultra-fast laser spectroscopy, dual polarisation interferometry, circular dichroism, multiangle light scattering, mass spectroscopy, small angle scattering, single particle analysis, electron paramagnetic resonance and powder difractometry.
