Abstract:

Synthetic adjuvants are one of the emerging trends in vaccine research. In an effort to develop a new class of vaccine adjuvant, we designed a focused library of synthetic glycophospholipids as immune potentiators. The in vitro preliminary evaluation has paved way for a lead molecule which was taken up for detailed in vivo adjuvant studies with a model antigen ovalbumin. Efficacy of a vaccine is based on stimulating both innate immunity and adaptive immunity. Therefore the antigen specific IgG response and its isotypes (IgG1 and IgG2a) were evaluated, followed by cytokines for humoral and cell-based immune responses were studied. Immunophenotyping of cells were also quantified proving the efficacy of glyco-lipid analogue as more desirable adjuvant.
