Abstract:

The manufacture of food products and dietary supplements using natural food pigments and other different natural compounds has been attracted attention in the field of modern food industry. One of the necessary and interesting natural compounds are carotenoids which show strong antioxidant and immunomodulation activities and may prevent degenerative diseases as well. The present research concerns the development and validation of a new, rapid, modern, effective and selective HPLC method for determination of carotenoids ?? beta-carotene and lycopene in organic extracts obtained from tomato, tangerine and orange agro industrial waste materials using extraction techniques-supercritical fluid and sequential ultrasonication-assisted extractions. The method was developed using RP-18 end-capped LiChroCART 4 x 250mm, 5μm column. The method was validated with respect to robustness, system suitability test, specificity, linearity-range, accuracy, precision, limit of detection (LOD) and quantitation (LOQ). The LOD and the LOQ are 0.081μg/mL and 0.041μg/mL for beta-carotene, 0.034μg/mL and 0.085μg/mL for lycopene, respectively. The content of each carotenoid per 1 g of dried local agro industrial waste material varies for beta-carotene 0.290-10.030 μg (tomato peel), 0.445-3.972 μg (tangerine peel), and 0.833-2.455 μg (orange peel) and for lycopene 0.418-143.79 μg (tomato peel), 0.051-179.988 μg (tangerine peel), 0.091-0.114 μg (orange peel).
