Development of manganese-quercetin complex for molecular imaging of cancer using MRI

Abstract :

It is well known that quercetin a plant-derived flavonoids exhibits various biological activities such as anti-oxidants, anti-cancer and anti-inflammatory. In particular for anti-cancer activity, quercetin was mediated cell cycle arrest and induction many cancer cell lines undergoing apoptosis through mitochondrial dependent pathway. In order to determine the tissular concentration and its pharmacokinetics by using molecular imaging quercetin was selected as ligand to form complex with Mn. For this talk will emphasize on the synthesis, spectroscopic properties and their magnetic susceptibility. The 1 mole of manganese and 2 mole of quercetin complex were presented the characteristic bands of absorption and emission at 440-485 nm and 540-585 nm, respectively. The Mn-quercetin complex enhanced the relaxation time of proton both T1 and T2. The cellular distribution of the complex can be studied by using fluorescent microscope. The accumulation of manganese-quercetin complex through green fluorescent in GLC4 cancer cell was found increased as a function of concentration. Mn-quercetin complex was found distribution in brain, liver, kidneys, bladder in normal rat and in addition found in the expected cancer tissues of rat bearing cancer cells. We proposed that the manganese-quercetin complex MR-imaging is a suitable technique for determining the tissular concentration of quercetin and also could be considered as a specific targeted contrast agent for cancer imaging.

Nathupakorn Dechsupa has completed his Ph.D at the age of 31 years from Chiang Mai University. He is a navigator for development of molecular imaging products (ND-MIPs) at the Center of Excellence for Molecular Imaging (CEMI), Chiang Mai University.
