Abstract:

The objective of present study is to prepare buccoadhesive film of Tri-metazidine di hydrochloride (TH-10mg), TH is a antianginal drug for the treatment of angina pectoris. Buccal films were prepared using different mucoadhesive polymers by solvent casing technique. The prepared films were characterized for physical appearance and surface texture, weight uniformity, thickness, folding endurance, swelling index, surface pH, drug content uniformity, tensile strength, and drug diffusion study. Formulations were subjected to weight uniformity, tensile strength, swelling studies, folding endurance, drug content shows satisfactory results. The percentage swelling index values were found to be increased with increasing the concentration of the polymer. The drug diffusion from the films was in the range of 61.93% to 91.23% till 8hrs. Formulations F1 and F4 shows the release from 91.23%, 84.03% respectively. Release of TH from the prepared films followed zero order kinetics and the mechanism was found to be non-Fickians diffusion and followed anomalous release. Hence the finding of this study indicated that the buccal films prolonged the TH release with a near zero order fashion. The moderate swelling, convenient resident time will lead to improve the bioavailability to a greater extend and better therapeutic efficacy of TH.
