The production of transgenic cells of plants, fungi and bacteria is a routine process. Nowadays, it is possible to insert into cells genes from plants, fungi, viruses, bacteria and even animals. Genetic transformation requires penetration of the transgene through the cell wall, a process facilitated by biological or physical methods. Efficient transformation techniques to control gene expression are needed to increase production levels and to ensure product quality. This is not trivial for numerous cells that could produce unwanted consequences. The recent discovery of shock wave-mediated genetic transformation of cells could be a solution to this problem. The goal of this presentation is to review the state of the art of the physical methods used for genetic transformation and to describe some of the basic physics and molecular biology behind them. It will give a general panorama of the traditional physical genetic transformation methods like DNA transfusion, electroporation, electrofusion, biolistics, agitation with glass beads, vacuum infiltration, silicon carbide whisker, laser microbeams, microinjection, macroinjection, and the newly promising technique of shock wave-mediated genetic transformation of cells.
Ana Leonor Rivera has completed her PhD on Sciences (Physics) from Universidad Nacional Aut?noma de M?xico (UNAM), made Postdoctoral studies from Institute of Nuclear Sciences, UNAM and was an Associate Member of the International Centre for Theoretical Physics (ICTP) in Trieste, Italy. She has been recognized as member of the Mexican National System of Researchers. She works as a full time Researcher of Center for Applied Physics and Advanced Technology, UNAM. She has published more than 30 papers in reputed journals, 5 chapters in books, and has been memory compiler of 7 academic events. Her research expertise is in the analysis of physical and biological signals using various mathematical methods and physical methods for genetic transformation.
