Pathway based omics data integration for identification of Cyclosporin A kidney toxicity related molecular mechanism

Abstract :

Cyclospoin A is widely used imunosuppressant drug for the prevention of rejection in organ transplantation. Nevertheless, it has adverse effects associated with molecular mechanism of action in diverse organs, such as kidney, nervous system, liver, blood, and intestine. To identify the molecular mechanism of drug toxicity, omics data is frequently used, so, in this study, we intended to know molecular mechanism of Cyclosproin A toxicity in kidney through omics data integration. It is not sufficient to explain the diverse toxicities of Cyclospoin A used only one type of data set , but data integration study improves the understanding of molecular mechanisms by complementary to each data sets. Therefore, we have performed a comprehensive characterization of molecular changes for rat in vivo model using transcriptomics, proteomics, and metabolomics analysis as it undergoes a Cyclosporin A administrated condition, and integrated three types of data. Transcriptomics, proteiomics, and metabolomics data got from microarray, ESI-MS/MS, and LC/ MS, respectively. We found significant molecular markers and pathways through over-representation and enrichment analysis in each data sets. After the comparison with the histopathology result, we could account the cholinergic synapse pathway related molecular changes with integrated molecular signatures. This study suggests that data integration method helps understanding the toxicity of drugs.
