To establish an HPLC method to determine the content of rhein,emodin,chrysophanol in Dongle plaster.Methods:Phenomenex Luna C18(250 mm×4.6 mm,5 μm)column was employed with methanol-0.1% phosphoric acid(85∶15)as mobile phase,the flow rate was 1.0 mL·min-1.Compounds was determined at λex 442 nm and λem 514 nm by fluorescence detector.Results:The contents of three components could be simultaneously determined;The calibration curves were linear in the ranges of 0.2752-27.52 μg·mL-1(r=0.9999)for rhein,0.2688-26.88 μg·mL-1(r=0.9996)for emodin,0.2560-25.60 μg·mL-1(r=0.9995)for chrysophanol;The average recoveries(n=3)of rhein,emodin,and chrysophanol were 96.3%(RSD=2.4%),97.6%(RSD=2.9%),99.0%(RSD=1.9%),respectively.Conclusion:The method is accurate with good reproducibility and strong specificity.It is suitable for determination of three compounds in Dongle plaster.
