Abstract A rapid method was developed to analyze bacterial behavioral responses to chemical stimuli. Digital image processing was used to detect the accumulation of bacteria at the mouth of a capillary containing an attractant. The accumulation of bacteria was determined from the total number of cells near the mouth of the capillary per videotape frame. This method was applied to measure the chemotactic response of Pseudomonas aeruginosa cells to serine, with results similar to those obtained by the classical capillary plating assay. The videotape method is much less time-consuming and makes it possible to assess the bacterial response to an attractant within a few minutes.
