Abstract The amount of urate depends on the balance between production and excretion. Although only about 10\% of hyperuricemia results from overproduction of urate, purine biosynthesis has substantial impacts on the pathogenesis of hyperuricemia. Here, regulation of purine biosynthesis pathway and relevant mechanisms are discussed. Genetic mechanisms for urate overproduction, dietary effects on urate amount and new drug therapy inhibiting urate production are also discussed.
