complimentary strand of molecule by addition of nucleotide bases with phosphodiester bond between 5’C of one nucleotide with 3’O of another nucleotide. In this study we proposed a chemoinformatics approach for synthesis of DNA polymer by fluorescent dye labelled nucleotide with a labile allyl group blocker at 3’O. The addition was read by fluorescent emission signal and the blocker stopped the further addition of nucleotide for the moment the addition was recorded, thereafter the blocker is cleaved by palladium catalyzed deallylation for further addition of fluorescent nucleotide on the complimentary strand of DNA molecule. This cycle continued for the synthesis of nucleotide bases on the complimentary strand. This method provides the best read length for target regions of the genome. The chemical mechanism of synthesis of allyl linker to the nucleotide, fluorescent dye labelled nucleotide, addition of nucleotide, deallylation of nucleotide and DNA polymer synthesis for reading the nucleotide sequence was elaborately investigated in this study.
