A reverse phase high performance liquid chromatographic method has been developed for the simultaneous estimation of penbutolol sulfate and hydrochlorothiazide in tablets. A Chromegabond M C 18 column with a mobile phase of 50 mM Na2HPO4: acetonitrile (60:40 v/v) adjusted to pH 3.5 with orthophosphoric acid was used. The flow rate was 1 mL/min and effluent was monitored at 220 nm. Samples as low as 278 ng/ml of penbutolol sulfate and40 ng/ml of hydrochlorothiazide can be estimated. Propranolol HCI was used as internal standard, and the order of elution was hydrochlorothiazide (HTZ) : propranolol : penbutolol (PEN) (3.43 : 5.57 : 10.76 minutes,respectively).
