| Figure 2: PQIP inhibits IGF1R signaling and induces apoptosis. Quiescent GEO cells were treated with 1μM PQIP (A) or 10μM LY294002 (B) for 24 hours, whole cell lysates were resolved by SDS-PAGE, and immunoblotted with different antibodies as indicated. Anti-actin polyclonal antibody was used as a loading control. GEO cells were seeded at 8,000 cells/well in 96-well plates and allowed to adhere overnight. The next day, cells were treated with different concentrations of either PQIP (A), or LY294002 (B). DNA fragmentation assay was performed and absorbance was measured at 405 nm (mean ± s.d.; n=3). |
