Figure 7: Quantification of positive and negative strand dengue viral RNA within the cells. Huh-7 cells were infected with DENV2 at m.o.i. of 1 in the presence and absence of emetine. The infected cells were then collected at different time points and subjected to real-time RT-PCR assay for quantification for positive and negative-sense DENV2 RNA species. Amounts of positive and negative-sense RNA per million cells are shown. The data are obtained from duplicate set of experiments. Emetine- treated cells exhibited reduction of positive-strand viral RNA by 1000-fold and negative-strand viral RNA by 10,000-fold when compared against 0.1 % DMSO non-treated, DENV2-infected cells at 24 h.p.i. The level of positive-strand and negative- strand of dengue RNA also remained significantly lower at later time point of 48 h.p.i. and 72 h.p.i.
