| Figure 3: Effect of T7-INcwt/mut on HIV-1 release and maturation. 293T cells were co-transfected with pNL4.3-GFP provirus and T7-INcwt, T7-INc215, 9AA or T7- INc240, 4AE expressing plasmid. After 48 h, virus-containing supernatant was collected and pelleted by ultracentrifugation. A: Viral production from transfected cells was monitored by measuring of HIV-1 p24gag antigen using the p24 ELISA assay. B: The virus particles were lysed and immunoprecipitated with anti-HIV-1 antibody and analyzed by western blot with mouse anti-p24 antibody (Upper panel). Levels of Pr55gag processing were calculated as the ratio of p55gag over p24gag, as quantified by laser densitometry (Lower panel). All results are representative of two independent experiments. |
