Mean Ct value ± SD1 for assays performed on 6 different days. Where a sample had no Ct value (i.e., Ct = 0), the number of days in which it had a Ct value (Ct = >0) out
of the six days it was tested is given in brackets2.
In Blue: denotes mean Ct values that are ≥3 more than expected.
In green: denotes unexpected false negative(s).
In magenta: denotes sensitivity of real-time RT-PCR for ISAV is below that of most laboratories.
In red: denotes poor performing laboratory based on having more than two days (a) with mean Ct values that are ≥3 more than expected, and/or (b) with unexpected
false negative(s)
, and/or (c) unable to detect a virus titer of 103 TCID50/ml.
General Observations:
1. Cut off Ct values range between≥40 and ≥35 = negative.
2. No lab had a false positive result.
3. Lowest amount of virus titer detected by most laboratories is 103 TCID50/ml.
4. The best performing laboratories are: Lab B and Lab E.
Table 3: Overall performance (inter-lab and inter-assay) of real-time RT-PCR for ISAV with TaqMan segment 8 probe (Snow et al. 2006) in Ring Test Phase II.
