Figure 4: Quantitative reverse transcription-polymerase chain reaction (qRT-PCR) analysis of expression of Insulin, Glut2, Nkx6.1 and Nkx2.2. Total RNA was extracted from BM-MSCs at the end of 3 steps of differentiation (S0, S2 and S3) in two different groups; BM-MSCs with and without co-culture. qRT-PCR detected the up-regulation of mRNA expression for insulin, Glut-2 and Nkx2.2 in last step differentiation comparing to step 0 and step 2. But the up-regulation of mRNA expression of Nkx6.1 did not detect in step 3 of any groups. For each sample, relative expression levels were normalized to undifferentiated GAPDH and undifferentiated BM-MSCs served as a negative control.
