Total protein extract (50 μg) from each biopsy was spiked with 30 μg of 13C6-Lysine labeled SILAC mouse muscle extract and processed using the pre- digestion spike-in approach (see Figure 1 and Methods). Intensity ratios of labeled and unlabeled peptide pairs were used to determine the absolute amount of dystrophin in each muscle extract.
