Abstract
Carbon dot (CD) has emerged as a potential tool to improve cancer treatment. In this study, we report a novel and facile method for large scale synthesis of highly fluorescent green carbon dots (G-dots) utilizing tea industry waste and its utilization as an efficient imaging and drug delivery platform for the purpose of targeted anticancer therapy. Despite large efforts to generate new platinum-based drugs and extensive preclinical testing, no drug superior to the already approved platinum complexes could be established. Inspite of having excellent anticancer efficacy, the nephrotoxicity and other toxicity issues restricted the use of platinum based drug. In this work, we have explored the drug carrying capacity of the prepared G-dots for targeted delivery of cisplatin with the aim of increase in the therapeutic index and decrease in the toxicity issues. We have used folic acid as navigational molecule due to its high level expression in most of the cancer cells. Folic acid and cisplatin were anchored with the G-dots (T-tags) through EDC/NHS coupling. T-dots; (0.1 mg/mL) induced death in cancer cells (HepG2 and MCF-7) in vitro at a higher rate than that of only cisplatin while the bare G-dot found to be non-toxic to the cell line tested. In vivo mice experiment showed enhanced anticancer efficacy of T-dots at 0.1 mg/ml in Dalton Lymphoma Ascitis (DLA) tumour and in DLA solid tumour model than that of the equivalent amount of free cisplatin, indicating that the loaded cisplatin retains its pharmaceutical activity.
