The seed and leaf protein content was estimated by adopting the methodology. 50 mg of oven dried black gram seeds and leaves were ground with the addition of 1 ml of 5% trichloroacetic acid. The absorbance was read at 660 nm using spectrophotometer. The total protein content was calculated by comparing the absorbance of each sample with a calibration curve plotted by taking known graded concentration of bovin serum albumin.
