Proliferation Assay

The proliferation assay was set up on a restimulation day for the MBP-specific T cell cultures or on the day of plating for the primary culture. The assay was performed in 96-well plates (Corning, Wilmington, NC) with 8-9A—104 MBP-specific T cells per well and a 6- to 10-fold excess of irradiated (3,000) isogenic speloncytes as antigen presenting cells per well.
