Numerous reports suggest that Aò aggregation,namely the formation of Aò oligomers, is an early event precedingother pathological changes in AD. Aò is physiologically generatedfrom the cleavage of amyloid precursor protein by ò- and ó-secretasesin the endoplasmic reticulum (ER) and trans-Golgi network or in thelate endosome, then secreted into the extracellular space.
