Topical Nutlin-3

(-)-Nutlin-3 (nutlin) is a cell-permeable antagonist of the murine double minute (Mdm2) p53 binding protein. Nutlin activates p53 by inhibiting the Mdm2-p53 interaction and is under development for the treatment and prevention of cancer. Nutlin increases apoptosis in many types of cells, but this effect in not universal. For example nutlin blocks UV-induced apoptosis in the osteosarcoma cell line U2OS and in keratinocytes via a pathway involving p21cip/ WAF and kinase JNK. Apoptosis in keratinocytes is mainly caused by cyclobutane pyrimidine dimers (CPD).
