Chemoproteomic approach based target deconvolution based on classical drug affinity chromatography has been successfully used in identification of molecular target of immunosuppressants and inhibitors of histone deacetylation. Protein kinases are major therapeutic targets and their involvement in cancer and inflammation has been well explored. Although several successful cancer drugs are associated with the well-defined protein kinase target profiles, such as Imanitib or Dasatinib, several off-targets have been identified for these drugs. The use of single immobilized kinase inhibitors allows the capturing of specific target proteins as well as off-targets.
