Brain sections were incubated with dihydroethidium (DHE) (stock solution, 1 mM in dimethyl sulfoxide, diluted to 5 μm with PBS, Molecular Probes, Eugene, OR, USA) in a humidified chamber, at 37°C for 30 minutes. Coronal brain sections were cut into 40 μm. A set of brain sections was processed for immunohistochemistry with polyclonal rabbit antimyeloperoxidase antibody (1:200, Thermo Fisher Scientific, Fremont, CA, USA) at 4°C overnight. Body weight measurements and neurologic evaluations were assessed at 72 hours after the ischemiaâreperfusion insult by one observer unaware of treatment protocol.
