RAPD analysis of brinjal (Solanum melongena L) genotypes for polymorphism associated with bacterial wilt resistance

Abstract
Bacterial wilt disease caused by Ralstonia solanacearum (Smith) Yabuuchi et al is one of the major problems for brinjal cultivation all over the world.Use of resistant varieties eliminates the excessive use of poisonous chemicals in controlling the disease which inturn reduces environmental pollution caused by them.Surya is a bacterial wilt resistant variety of brinjal developed by Kerala Agricultural University. The source of resistance in this variety could be used to develop transgenics not only in brinjal but also in other susceptible genotypes. The present study was undertaken to develop a molecular marker linked to the bacterial wilt resistance gene in Surya .RAPD analysis of the same was done with Pusa Purple Long, a susceptible variety released from IARI. DNA isolation was done using Rogers and Bendich (1994) method. The purity of DNA was further analysed using Nanodrop Spectrophotometry. Good quality DNA of UV absorbance ratio (A260/A280) in the range of 1.8- 2 was used for study. Sixty random decamer primers from Operon series were used for PCR amplification to detect polymorphism between selected varieties. The primer OPS 17 has given a polymorphic band of 1375 bp in resistant variety.

Somya .P.P did her BSc Agriculture from College of Agriculture,Padannakkad. Now she is pursuing her Masters degree in Agricultural Biotechnology from College of Horticulture, Thrissur under the guidance of Dr .P.A.Valsala.
