 Biography :

Sambamurthys research aims to understand the basis of neurodeneration in age-related neurodegenerative diseases. We are using multiple approaches in several disease models to systematically uncover the common pathways in neurodegeneration and to identify methods to prevent or treat the process. A key focus on the lab has been Alzheimer’s disease (AD). In this disease, there is a characteristic deposition of the 42-residue amyloid β protein (Aβ42) demonstrating the failure of protein homeostasis in the brain.  Similar protein accumulation is characteristic of a number of other degenerative diseases in the CNS such as age-related macular degeneration (ARMD) as well as somatic tissues as in islet amyloid. The general assumption has been that the neurodegeneration is caused by specific toxicity of the accumulated protein aggregates.  Interestingly cholesterol has been identified as a risk factor in AD and also in ARMD. Both diseases show extracellular deposits of amyloid. Our studies are evaluating the role of amyloid and the secretases in AD as well as retinal generation to understand common mechanisms of degeneration. We hope and expect that the detailed evaluation of the common pathways of degeneration in AD and forms of retinal degeneration will identify ways to prevent and treat these devastating diseases.

Dr. Joseph Quinn is an Associate Professor of Neurology at Oregon Health and Sciences University and a staff neurologist at the Portland VA Medical Center.  He completed his undergraduate studies in Biochemical Sciences at Harvard University, his MD at University of Southern California, his internship at LA County Hospital, and his neurology residency at Oregon Health and Sciences University. He has been on the faculty in Oregon since 1994. With grant support from the NIH and from the VA, he has conducted laboratory and clinical research on a number of such candidate therapies for Alzheimer’s disease, including ginkgo biloba, melatonin, lipoic acid, ibuprofen, resveratrol, coenzyme Q, copper chelation, and the omega 3 fatty acid DHA. His research is described as “translational”, which means Dr Quinn has no original ideas but instead attempts to bring established ideas from the laboratory to clinical trials. In some cases, such as copper chelation therapy, this means conducting animal studies to determine if clinical trials are warranted. In other cases, such as ibuprofen, this means conducting novel pilot clinical trials utilizing surrogate markers of disease in order to determine if a large scale clinical trial is warranted.  And in yet other cases, such as a national multi-center trial of DHA , this means designing and supervising a full scale clinical trial based on preclinical animal studies conducted and reported by other investigators. The specific positions held by Dr Quinn to support these research efforts include Biomarker Core Leader, Associate Clinical Core Leader, and Clinical Trials director for the NIA-funded Oregon Aging and Alzheimer’s disease center. Dr Quinn is also co-director of the Portland Parkinson’s Disease Research, Education, and Clinical Care Center.

Cara J. Westmark
Department of Pathology & Laboratory Medicine
University of Wisconsin-Madison
Waisman Center for Developmental Disabilities
USA
Tel. (608) 262-9730

 Biography :

Dr. Cara Westmark is an Associate Scientist at the Waisman Center for Developmental Disabilities at the University of Wisconsin-Madison. Her current research examines the role of genetic and environmental factors on the expression of amyloid precursor protein (APP) and beta-amyloid (A beta) in transgenic mouse models of neurological disorders. She has demonstrated that APP/ A beta levels are altered in Fragile X syndrome and that many FXS phenotypes can be rescued in a mouse model of the disorder by genetically reducing APP/ A beta. These data suggest that therapies studied for the treatment of Alzheimer’s disease (AD) may be beneficial in the treatment of FXS. She has also demonstrated a strong association between APP/ A beta over-expression and lower seizure threshold, which has important implications for the treatment of seizure-related disorders such as AD, FXS and Down syndrome. Her most recent work has demonstrated a strong association between dietary phytoestrogen consumption, elevated A beta and reduced seizure threshold.
