Dr. Wayne Grant Carter received his Honours degree in Biochemistry with Nutrition from the University of Southampton. He then completed a PhD at the University of Southampton studying the molecular signalling cascade elicited by insulin. His PhD studies encompassed the design and synthesis of peptide targets for insulin stimulated kinases and their utilisation to dissect signalling events downstream of insulin binding to its receptor He purified peptide targets by reversephase high performance liquid chromatography RPHPLC and assayed signalling kinases utilising radiolabelled phosphorus to determine phosphorylation sites by both radiosequencing and phosphopeptide mapping eg Carter et al 1995 1996 Subsequent postdoctoral studies at the Babraham Institute Cambridge; Imperial College London; University of California at Irvine; and University of Oxford were also directed toward the regulation of cellular signalling mechanisms through posttranslational modification of proteins and how these influence inflammation and homeostasis Dr Carter then spent one year working for Mobious Genomics Exeter assisting with the biophysical development of novel DNA sequencing devices before he joined the staff at the University of Nottingham where he is currently a Principal Investigator and Lecturer for a new course in Healthcare Sciences founded at the University of Nottingham site within the Royal Derby Hospital Derby His previous and current research pioneered the use of proteomic techniques coupled to novel imaging of radiolabels to detect and purify proteins that are modified posttranslationally eg Vigneswara et al 2006; Carter et al 2007; Tarhoni et al 2008; Carter Aswad 2008; Kharbanda et al 2009; Carter et al 2010 He has received external grant funding for his academic studies from research funding bodies that include the Wellcome Trust UK and the National Institutes of Health USA and also from commercial sources that include Syngenta UK His professional affiliations include The Physiological Society the Royal Society of Chemistry the Biochemical Society the British Toxicological Society the British Neuroscience Association and the American College of Sports Medicine He has peerreviewed manuscripts for numerous academic journals including: FEBS Letters Brain Research Environmental Toxicology Biochemica et Biophysica Acta American Journal of Physiology Journal of Chromatography B International Journal of Biomedical Science World Journal of Cardiology and Toxicological Sciences and is currently an Editorial Board member for the World Journal of Biological Chemistry as well as Executive Editor for Europe for the Journal of Chromatography Separation Techniques

Saliva is well known to play a major role in maintaining oral health conditions by interfering with the adverse affects of oral pathogens. My research interests focus on the structure-function relationships of salivary proteins. Most important in this host protection by saliva is the capacity of several salivary proteins to protect oral hard and soft tissues. More recent studies with pure salivary proteins isolated from sterile glandular secretions have shown that functional domains representing short segments of the native proteins also exhibit antimicrobial and mineral homeostasis characteristics. Once these native proteins derived from the glandular secretions enter the oral cavity they are exposed to a host of enzymes including proteases. My projects are specifically geared towards an understanding of the proteolytic fragmentation of these glandular proteins using proteomics approaches for their structural characterization and specific bioassays to evaluate the effects of proteolysis on the functional spectrum of these salivary macromolecules. .

After finishing his PhD in 1994 he spent two years in Department of Genetics at Yale University as a postdoctoral fellow working on research problems in metabolic diseases of mitochondrial origin. In 1999 he joined a research group at the Mayo Clinic working on disease mechanisms in Friedreich’s ataxia. In 2002 Dr. Adamec accepted the invitation of the founders to assume the position of Senior Scientist at Beyond Genomics a biopharmaceutical company founded to discover and develop new drugs with Systems Biology approaches. Currently he is working as Research Associate Professor and a Lead Scientist for Proteomics and Metabolomics at the Bindley Bioscience Center at Purdue University. Dr. Adamec has authored more than 40 scientific journal publications. He is also author of three book chapters multiple patents and associated editor Journal of integrated OMICS.

Dr. Timms conducts basic and translational cancer research through the integration of proteomic, genomic and molecular biology techniques. His group has established several proteomic platforms and applied these for profiling of clinical samples and cell models with the aim of improving proteomic coverage, sensitivity and quantitative accuracy. Proteomic platforms and their applications include: high-throughput protein/peptide extraction and MALDI-TOF MS profiling of unique serum collections for ovarian, breast, pancreatic and biliary tract cancer biomarker discovery; 2D-DIGE/MS-based expression profiling applied to tissue, cell model and serum protein fractions to identify candidate markers of breast and ovarian cancer, intermediates in growth factor signalling and proteins involved in tumour suppression and stress responses; incorporation of stable isotopic and isobaric labelling methods (SILAC, TMT), 2D-LC separations and phosphopeptide enrichment into proteomic workflows for biomarker discovery and functional characterisation. These advanced methods are being employed on several projects aimed at cancer biomarker discovery and validation and the characterisation of the molecular and cellular events associated with tumourigenesis.

I have extensive expertise in protein chemistry and glycobiology and have been working on the basic and translational research of galectins. In my postdoctoral research, I studied on the structurefunction aspects of the lectins, particularly galectins from vertebrates and invertebrates. The structurefunction studies enabled me to predict the type of endogenous ligands for a particular galectin. As a coinvestigator on a NIHfunded grant, I identified and characterized the zebrafish galectin repertoire, which showed remarkable structural similarities with mammalian galectins. The two proto type galectins, Drgal1L2 and Drgal1L4 are found specifically expressed in the notochord and are involved in muscle cell development including heart. As PI on three federal NIH and DOD and one nonfederal university grants, I have been studying on human galectins, particularly on application and manipulation of galectin3 for the diagnosis and prevention of cancers. In addition, I successfully administered the projects including research protections, budget, and collaboration and produced peerreviewed publications. In summary, I have a demonstrated record of successful and productive research projects in an area of lectincarbohydrate interactions.

My research interest has been in the area of basic and translational research of the carbohydrate-binding proteins (called lectins). Particularly, I am interested in structure, function and regulation of galectins (a family of beta-galactoside-binding lectins) and their interactions with the carbohydrates that mediate cell-cell and cell-extracellular matrix (ECM) interactions during normal and cancer development such as prostate and breast cancers. The prostate cancer cells differentially express several members of galectins, which alter normal cell-cell and cell-ECM interactions during cancer development. We have discovered a novel isoform of galectin-8 that may be relevant to prostate cancer cell proliferation. We have demonstrated cytosine methylation in galectin promoter in cancerous prostate, which may account for the differential expression of galectins during cancer development. Differential expression of galectin repertoire and identification of cytosine methylation of galectin gene promoters in normal and tumor prostate tissues have enabled us to develop a methylation-specific PCR based sensitive assay for early diagnosis of prostate cancer in biological fluids such as serum and urine. In another project, natural carbohydrate inhibitors of galectin are being employed to prevent breast cancer metastasis.

To design and synthesize novel carbohydrate-based conjugates mimicking the HIV glycan shield is an HIV vaccine strategy for eliciting 2G12-like neutralizing antibody. Dr. Huang’s another project focuses on glycophosphatidylinositol (GPI)-based vaccine studies against GPI-related infectious diseases, such as malaria, trypanosome, etc.
