The goal of this proposal is to determine the mechanism by which topical application of the opioid antagonist naltrexone accelerates the healing of skin wounds in Type 1 diabetic mice The basis of this proposal stems from our long time expertise in opioids cell proliferation wound healing and diabetes In 1983 using a paradigm of intermittent and continuous opioid receptor blockade with naltrexone NTX we Drs Zagon and McLaughlin discovered that endogenous opioids and opioid receptors previously thought to be related only to neural cells and neurotransmission serve as determinants of cell proliferation This has led us on a journey of discovering the opioid peptide [Met5]

 Research Interest :

One direction of our research has been to understand the role of opioids in diabetes. It is well known that diabetics have an elevate level of enkephalins and problems with pain perception. We have connected the increased levels of opioids with delays in wound healing commonly occurring in diabetics. In grants sponsored by NIDDK, we have found that a topical application of NTX accelerates corneal wound healing in animals with Type 1 diabetes by disrupting the enhanced interactions of the OGF-OGFr axis in these animals. Introduction of sense or antisense of OGFr into the cornea by a gene gun revealed that the OGF-OGFr pathway is vital to regulating cell proliferation and wound healing. In the course of these experiments, we have devised a topical application of a cream of NTX that accelerates the closure of skin wounds in diabetics. NTX is known as a pure biological compound with little to no direct effects, and we hypothesize that NTX is accelerating wound healing of skin by blocking a critical opioid peptide-opioid receptor pathway. This grant application seeks to understand the underlying basis for these phenomena and tests our hypothesis in mice with Type 1 diabetes by employing knockout animals for classical and non-classical (OGFr) receptors and studying the absence of receptors on wound healing. In a third specific aim, we will use tissue culture of epithelial cells along with a wound model to integrate information in Specific Aims 1 and 2 with the cell and molecular biology of opioid action. In summary, we have pioneered in the use of NTX to upregulate wound healing, developed a topical NTX that is safe and efficacious for skin wound repair, have the reagents, tools, and experience to conduct the [-enkephalin - termed opioid growth factor (OGF) in view of its non-neural and neural distribution as well as its function in growth), and opioid receptor (OGFr) associated with cell proliferation. We have partnered with clinicians to translate our science from bench to bedside, and clinical trials have been published regarding the use of OGF - and low dose naltrexone (LDN) - for the treatment of cancer and autoimmune diseases showing safety and efficacy, and topical application of NTX is now being examined in a clinical trial sponsored by the Department of Defense by my co-investigator, Dr. Joseph Sassani. proposed research, demonstrated a record of successful and productive research projects in an area of high relevance to human health and disease, and we are addressing an important unmet medical needs.

Human T-cell leukemia virus type I (HTLV-I) was the first retrovirus shown to cause cancer in humans. The major viral oncoprotein, Tax, is a transcriptional regulator of viral and cellular gene expression, and interacts with many important cellular proteins that regulate cell cycle progression, and cellular responses to DNA damage. The ongoing studies in my lab encompass a broad-based effort to understand the molecular mechanisms of Tax function, and how these functions promote genomic instability and cellular transformation.

Emanuela Taioli MD PhD is Professor of the Department of Epidemiology and Biostatistics and Professor of Hematology and Oncology within the School of Medicine at SUNY Downstate From 2005 to 2008 she was Director of the Division of Cancer Prevention and Population Science at the University of Pittsburgh Cancer Institute where she also held the Arnold Palmer Endowed Chair in cancer Prevention In this capacity she has developed a successful partnership with Hampton University under NIH funding to teach Epidemiology to undergraduate students From 20022004 Dr Taioli was a technical advisor to the Italian Ministry of Health In this role she served as a member of the European High Group of Reflection on Patients Mobility Previously Dr Taioli was Director of the Unit of Molecular and Genetic Epidemiology at the main University Hospital in Milano Italy and the PI of the GSEC project an International Study on Genetic Susceptibility to Environmental Carcinogens Started in 1997 and funded by the European Commission for Research GSEC is a pooled analysis of individual epidemiologic and genetic data including over 200000 subjects Dr Taioli was also an assistant professor in the Department of Environmental Medicine at New York University In this role she conducted studies on genetic susceptibility to environmental factors in lung and breast cancer and differences in estrogen metabolism with ethnicity in womenDr Taioli obtained her Medical Degree from the University of Milano where she also completed her Residency in Cardiology She obtained an MS and a PhD in Epidemiology from Columbia University Dr Taioli also worked under a NATO fellowship at the American Health Foundation in New YorkDr Taioli is also the coauthor of over 260 peer reviewed papers

Following a BSc in Chemistry and Biology VD pursued a PhD in Biochemistry at the University of Western Ontario His interests in receptors and signalling lead him to pursue a postdoctoral position at the Wellcome Research Laboratories in Research Triangle Park NC with Drs Pedro Cuatrecasas and Steven Jacobs Returning to Canada he joined the Biomedical Research Centre at UBC in Vancouver to begin work on signalling via cytokine receptors Since joining UBC as a faculty member in the Dept of Medicine he has progressed through the ranks and is currently appointed as Professor He also serves as director of the UBC Experimental Medicine Graduate Program with almost 200 students under the supervision of faculty members in a variety of clinical departments

His research interests have focused on various aspects of signal transduction, with initial emphasis on how the PI 3-kinase pathway was involved in cell survival. This work has also been investigating the control of the life and death balance in cells via control of BCL-2 family proteins. More recently, a number of alternative functions of BCL-2 family proteins have been characterized, including involvement in regulation of cell cycle and involvement in DNA damage response. Being a basic researcher in a clinical department has offered opportunities for several collaborations with clinician-scientists, leading to projects in a diverse set areas including atherosclerosis, tendon damage, inflammation and fibrosis in the respiratory field, multiple sclerosis and of course, cancer.

Cancer is a nondiscriminatory disease afflicting millions of Americans annually One hallmark trait of many cancer cells irrespective of tissue type is the ability of cancer cells to grow faster than normal cells RNA polymerase III RNA pol III is an enzyme complex that catalyzes the transcription of many of the small structural RNA molecules required for RNA processing U6 snRNA and translation tRNA Thus RNA pol III plays a key role in regulating the growth of a cell because it plays an important role in the regulation of protein synthesis and for a cell to grow or multiple proteins are needed RNA pol III products have been demonstrated to be specifically elevated in a variety of cancers Accurate initiation by RNA pol III requires a protein complex called TFIIIB implicating TFIIIBmediated transcription as an important determinant of the biosynthetic capacity of a cell TFIIIB is a target for repression by tumor suppressors proteins that inhibit cell growth and activation by oncogenes proteins that activate cell growth We are actively seeking to identify the mechanisms by which RNA pol III transcription becomes deregulated in breast cervical and prostate cancer Our long term goal is to use our findings to ultimately identify novel pharmacological targets for cancer drug development The tumor suppressor BRCA1 has been shown to be mutated in familiar breast cancer BRCA1 plays a role in DNA repair cell cycle regulation and programmed cell death apoptosis as well as other cellular activities My laboratory has recently demonstrated for the first time that BRCA1 inhibits RNA pol III transcription and we hypothesize that BRCA1 negatively regulates RNA pol III transcription through TFIIIB The longterm goal of this project is to elucidate the mechanisms by which BRCA1 regulates RNA pol III transcription through TFIIIBNutraceuticals and other naturally occurring pharmacologically active components have been gaining increasing popularity in the United States especially in medicine Scientists are trying to determine the molecular mechanisms of how these nutraceuticals work at the level of the cell One such compound is EGCG the major component of green tea Green tea has been demonstrated to inhibit the growth of a variety of cancer cells but the molecular mechanism of how EGCG inhibits cancer cell growth is not well established We do know that both EGCG and RNA pol III have the capacity to modulate cell proliferation Thus we hypothesized that EGCG could modulate RNA pol III transcription Our data demonstrates that EGCG can repress RNA pol III transcription in cervical prostate and breast carcinoma cells The long term goal of this project is to elucidate the molecular mechanisms governing the regulation of RNA pol III transcription by the green tea component EGCG to identify novel pharmacological targets for cancer therapy My laboratory is also examining the regulation of RNA pol III transcription by other bioactive compounds such as resveratrol red wine and genistein soy in cancer Through this research we hope to determine the mechanisms by which natural products inhibit cancer cell growth

The goal of the proposed research is to establish nanovectors as a gene/miRNA delivery platform to develop novel molecular therapies targeting cancer stem cells I have the expertise leadership and motivation necessary to successfully carry out the proposed work I have a broad background in cancer biology and nanobiotechnology with specific training and expertise in key research areas for this application including molecular cancer therapeutics microRNA nanovectors for gene delivery As a postdoctoral fellow at Stanford and Georgetown I carried out pioneer studies in designing novel nanovectors for tumortargeted gene delivery which eventually obtained multiple patents and FDAapproval for the firstinman clinical trials of nonviral targeted p53 gene therapy At the University of Michigan I expanded my research to develop novel nanovectors to deliver siRNA and miRNA based therapeutics as well as smallmolecule therapeutics As PI or coInvestigator on several previous NIH and DODfunded grants I laid the groundwork for the proposed research by developing antiCD44nanovectors that can target up to 66 cancer stem cells in vivo in a pancreatic tumor model and by establishing a collaborative network of experts in cancer stem cells miRNA and nanotechnology In addition I successfully administered the projects eg staffing research protections budget collaborated with other researchers and produced several peerreviewed publications from each project As a result of these previous experiences I am aware of the importance of frequent communication among project members and of constructing a realistic research plan timeline and budget The current application builds logically on my prior work on nanovectors and I have chosen coinvestigators Drs Simeone and Liu who provide additional expertise in cancer stem cells and human primary tumor models In summary I have a demonstrated record of successful and productive research projects relevant to the current proposal and my expertise and experience have prepared me to lead the proposed project

Molecular therapy targeting cancer and cancer stem cells, cancer biology, drug discovery and drug/gene delivery.Funded by multiple NIH and DOD grants, my lab is working on molecularly targeted cancer therapy and chemo/radiosensitization by modulating cell death signaling pathways, autophagy and apoptosis. We are exploring microRNAs as novel targets for cancer and cancer stem cells. We are currently using novel nanotechnology to develop nanovectors targeting cancer cells for siRNA/miRNA-based novel cancer therapeutics. We are also developing nanovectors targeting cancer stem cells and exploring novel molecular therapy for cancer stem cells.

The research of Dr. Frost’s laboratory is focused on understanding the effects of the stromal microenvironment of the breast on breast carcinogenesis and the progression of breast cancer, with the ultimate goal of identifying new targets for the prevention of invasive and metastatic disease. Current work includes the study of the role of Hedgehog signaling and Gli-mediated transcription in breast cancer, in both epithelial cells and fibroblasts. Because primary cilia are important in Hedgehog signaling, the presence and function of primary cilia in breast cancer and normal breast is also a topic of current interest.

After getting his PhD degree at University of Maryland School of Medicine Human Genetics Program in 1998 he joined the research fellow at National Institutes of Health where he trained cancer biology and pharmacology During his fellow Dr Baek found a protein NAG1 induced by nonsteroidal antiinflammatory drugs which are known chemopreventive drugs in colorectal cancer Dr Baek joined the faculty of The University of Tennessee College of Veterinary Medicine and pioneered defining the critical target genes in chemoprevention and understanding how anticancer compounds act upon these genes to influence cancer prevention or development Several chemopreventive target genes were identified and characterized in his laboratory including activating transcription factor ATF3 early growth response EGR1 NSAID activated gene NAG1 and other tumor suppressor proteins Interestingly these proteins were also induced by several dietary compounds and PPAR? ligands supporting their role in anticancer antiobesity and antiinflammation activities His findings have important consequences in the development of new drugs for the prevention of cancer since it may be possible to develop drugs that selectively increase NAG1 and others in tumors and they provide a mechanistic explanation for preventing tumor development He is currently an Associate Professor at The University of Tennessee College of Veterinary Medicine and has published more than 70 manuscripts on the molecular targets of several chemopreventive compounds including phytochemicals PPAR? ligands and NSAIDs on colorectal tumorigenesis

My laboratory is interested in the molecular and cellular pharmacology of anticancer drugs, such as drug resistance mechanisms and reversal of resistance study and new drug development.A promise of our research program is that progress in the chemotherapy of cancer will depend on rational combination of agents that reduce the function of plasma membrane transporters (ATP-binding cassette trasnporters) such as P-gp, ABCG2 and MRPs, and to enhance the actions of cytotoxic drugs. In collaboration with many researchers in USA, China, Japan, Australia and Egypt, studies in our laboratory focus on screening and identifying new inhibitors of ABC transporters by in vitro membrane vesicles transport assay system and cell based assay systems. Another future research direction is to study the role of MRP family on the detoxification of chemicals using knock out mouse models.

I am originally from Iowa and attended Iowa State University where I received a BS degree in Biochemistry Following that I then received my PhD degree in Biochemistry from The University of South Dakota School of Medicine The focus of my doctorate studies was DNA repair studies in Chlamydomonas rheinhardi I next completed two postdoctoral positions at Mt Sinai School of Medicine in New York City working first with Dr David Calhoun in Microbiology and then Dr Phyllis Shaw in Anatomy/Cell Biology I am currently an associate professor of Biochemistry at AT Still University in Kirksville Missouri I work with metastatic melanoma cells and am particularly interested in mechanisms of tumor cell migration and invasion by proteolytic enzymes

The proposed research has five major goals One is to confirm preliminary data indicating dramatic deficiency in expression of DNA repair proteins ERCC1 and Pms2 as frequent early events forming large field defects in progression to colon cancer in humans A second goal is to characterize the mechanisms by which the ERCC1 and/or Pms2 deficiencies arise A third goal is to determine the mechanistic consequences of ERCC1 and/or Pms2 deficienciesA fourth goal is to characterize our new unique dietaryrelated wildtype mouse model of colon cancer to relate it to progression to colon cancer in humans A fifth goal is to use this mouse model to determine the protective effects of dietary caffeic acid resveratrol phenethyl isothiocyanate PEITC found in cruciferous vegetables and curcumin in prevention of colon cancerI have long term demonstrated expertise and motivation to carry out the proposed work As early as 1991 using deoxycholate and Chinese hamster ovary cells deficient in ERCC1 mutant cells UV4 I proposed that bile acids have an etiologic role in colon cancer by causing DNA damage Since then I have been first author or coauthor on 41 additional articles on the roles of bile acids field defects and DNA repair enzymes in colon carcinogenesis as well as use of bile acids in mouse models of inflammation Barrett’s esophagus and promotion of colonic adenomas The current application builds logically on my prior work and my physician coPI is a research oriented physician scientist who is actively involved in elucidating factors involved in progression to colon cancer

Dr. Turners research program focuses on determining the impact of dietary constituents on regulatory processes that may protect against carcinogenesis and inflammation in the colon. Her lab is evaluating the effects of fiber sources and the specific phytochemicals contained within them on aspects of cellular proliferation or apoptosis, and microbial/epithelial cell interactions. The goal is to determine how these normal processes are being perturbed by chemical carcinogens, radiation or pro-inflammatory compounds, and how diet may mitigate the damage caused by them. Work conducted in the laboratory is currently funded by the USDA/CSREES through the Texas A&M University Vegetable and Fruit Improvement Center, the USDA/ARS, and the National Space Biomedical Research Institute.

 Biography :

Dr. Xiaoyan Jiang obtained a MD from Shanghai Second Medical University, Shanghai, China and a PhD (Molecular Biology) at McGill University, Montreal, Canada. She is now a Senior Scientist in the Terry Fox Laboratory of the BC Cancer Agency, an Associate Professor in the Department of Medical Genetics and an Associate Member in the Department of Medicine at the University of British Columbia.  She is also an Adjunct Professor at the Shanghai Institute of Medical Genetics of the Shanghai Jiao Tong University School of Medicine.  In 2005, she was awarded as a Michael Smith Foundation for Health Research Scholar.  She has presented at numerous international meetings, including a number of invited presentations, and is active as a research grant reviewer and an external (ad hoc) reviewer both in Canada and the USA. She is serving as an editorial member of several reputed journals and is a member of American Society of Hematology and International Society of Experimental Hematology since 1998.
 
Dr. Jiang’s research interests are focused on basic and translational leukemia research, particularly human chronic myeloid leukemia (CML).  The ultimate objective is to identify new, rationally designed, molecularly targeted therapies that will be more effective and less toxic than those presently available.  Recently, her research group has identified a novel AHI-1-BCR-ABL-JAK2 interaction complex that modulates BCR-ABL transforming activity and imatinib response of CML stem and progenitor cells.  Understanding the functions of this new interaction complex could lead to the development of new molecular targeted therapies.

Dr William Chi-shing Cho is a scientific officer at the Department of Clinical Oncology in Queen Elizabeth Hospital. He is a Chartered Scientist granted by the Science Council (UK) and a fellow member of several institutes, including the Institute of Biomedical Science (UK), Hong Kong Institute of Biomedical Science and Hong Kong Society for Molecular Diagnostic Sciences. Dr Cho has published over 100 papers and plenty of books covering cancer biomarkers, proteomics and microRNAs. He serves as the editor-in-chief, editor and associate editor of a number of international medical journals. Dr Cho is also an international grant reviewer of the Hope Funds for Cancer Research (USA), Cancer Research UK, Science Foundation (Ireland), National Medical Research Council (Singapore), and Academia Sinica Investigator Award (Taiwan).

 Biography :

Dr. Rui Amaral Mendes attained his DMD degree from the University of Porto in 2000. After initiating is doctoral studies at the University of Porto, he afterwards proceeded to the Vrije Universiteit Amsterdam where is currently completing his doctoral thesis on keratocystic odontogenic tumors of jaws. He was appointed Assistant Professor of Oral Medicine and Oncology of the Portuguese Catholic University in November 2009. Since then, he has also been working as Assistant reviewer of the V World Workshop on Oral Medicine – “Genetics/epigenetics of oral mucosal premalignancy” and he is also member of Research Interest Group "Head & Neck Oncology” of the International Association of Oral and Maxillofacial Surgeons. He is Fellow of the International Association of Oral and Maxillofacial Surgeons and Fellow of the International Academy of Oral Oncology.

Idriss M. Bennani-Baiti

 Biography :

Dr. Bennani-Baiti, a former Howard Hughes Scholar and Human Genetics Fellow at University of Pennsylvania School of Medicine, carried out research into the molecular mechanisms of gene regulation at the Memorial Sloan-Kettering Cancer Center. He later joined the faculty of St. Louis University School of Medicine wherein he continued this work. He currently leads several research programs at the Children′s Cancer Research Institute into the pathology of childhood cancers, with a particular interest in sarcomas and other mesenchyme-derived tumors. He is also interested in the deregulation of signaling pathways (e.g. p53, Notch) and miRNAs in cancer, in epigenetics and epigenomics, and in novel methods to inferring tumor biomarkers from Omic datasets. Dr. Bennani-Baiti is an active member of the American Association for Cancer Research, AACR Pediatric Cancer Working Group, Epigenetics Society, International Society for Systems Biology, and Society for Mathematical Biology.

Dr. Bennani-Baiti is interested in the mechanisms of tumor pathogenesis, as well as in tumor, metastasis, and prognosis markers, with a current focus in childhood cancers. He has recently developed bioinformatic and mathematical methods to uncovering genes significantly associated with particular disease states from small-size cohorts (which cannot be analyzed with conventional statistical methods for lack of statistical power), the only cohorts available for most childhood and adult cancers. Using such methods, he has for example discovered the first metastasis and prognosis molecular markers of Ewing′s sarcoma, a highly aggressive tumor of bone and soft tissues that afflicts mostly children and young adults. He is also interested in inferring signaling pathway status and connectivity directly from tumor specimen. He is also conducting investigations into the epigenetic mechanisms of tumorigenesis.
