 Research Interest :

Dr. Eldred\\\'s research for the past 15 years has examined the role of the gaseous neuromodulator nitric oxide (NO) in retinal signal transduction and ocular pathology. He has determined that nitric oxide is reciprocally related to many retinal neurotransmitters including GABA, glycine, acetylcholine, and dopamine in that NO modulates their release and they in turn modulate NO production. He has also shown that every cell type in the retina can produce NO. Using direct imaging of NO, he established that in retina NO is not freely diffusible. He is now examining the role that NO plays in the early neuronal cell death seen in diabetic retinopathy by focusing on the role that the signaling peptide adrenomedullin plays in producing the pathological increases in NO found in early diabetic retinopathy. He has established and localized all of the biochemical steps in this pathway and is now developing pharmacological interventions to inhibit the pathological activation of the adrenomedullin/NO signaling pathway. Since both NO and adrenomedullin are involved in much ocular pathology including glaucoma, ischemia, and uveitis, a better understanding of the adrenomedullin/NO signaling pathways will have broad clinical applicability.

His research interest is in the fields of endocrinology and metabolism for bone and calcium since 1971 years, and it has been developed in the aspects of endocrinology, biochemistry, nutrition, pharmacology and toxicology. Especially, he has an interesting for bone biology, osteoporosis prevention by nutritional factors, diabetic osteoporosis and intracellular signaling mechanism. He discovered two novel proteins and genes; the named regucalcin, a regulatory protein in intracellular signaling system, and RGPR-p117, a transcription factor. Regucalcin transgenic rats are generated and this rat has been known to induce osteoporosis and hyperlipidemia.

Dr Naila Rabbani graduated with a BSc in Biological and Medicinal Chemistry in 1998 and PhD in Disease Mechanisms and Therapeutics in 2001 at the University of Essex Colchester UK She pursued predoctoral research improving analytical methods to identify and quantify markers of protein damage by glycation and has maintained an interest in protein glycation in disease mechanisms and therapeutics She received Shaul Massry Prize for the best oral presentation at the 4th International Congress on Uremic Research and Toxicity in 2005 She was awarded a British Heart Foundation Intermediate Fellowship in 2006 for research on glycation of lipoproteins and became an assistant Professor in Experimental Systems Biology at the University of Warwick in 2007 and promoted to Associate Professor in 2010 She is Codirector – with Professor Paul J Thornalley of the Protein Damage and Systems Biology Research Group Warwick Medical School She employs gold standard stable isotopic dilution analysis mass spectrometry for comprehensive and quantitatively screening of amino acids and related glycated oxidised and nitrated derivatives in mammalian cell culture preclinical animal pharmacology and clinical studies – including working with several national and international collaborating research groups She has over 50 publications in peerreviewed journals and more than 65 abstracts/conference papers

Her current research focuses on investigations of damage to lipoproteins and influence of dietary bioactive compounds on lipoprotein synthesis, damage and metabolism - supported by the BHF and BBSRC. Other activities: (i) Study co-ordinator of a pilot-scale placebo-controlled, double blind clinical trial of high dose thiamine therapy in type 2 diabetic patients with microalbuminuria in Shaikh Zayed Hospital, Lahore, Pakistan. The outcome - published recently – showed reversal of early stage kidney disease and improvement of kidney function of all patients receiving thiamine. (ii)She is Editor of IMARS Highlights – the bimonthly newsletter of the International Maillard Reaction Society (IMARS). (iii)Founding co-editor of the glycation section of the journal Amino Acid (first issue will be published by the end of 2010)

Her research focused on investigating thiol/disulfide redox system and reactive oxygen/nitrogen species-controlled signaling mechanisms in association with atherosclerosis development. During the last several years she has developed a new technique for redox proteomics by redox ICAT analysis combined with mass spectrometry, which identifies redox-sensitive proteins and their associated mechanisms. Her current research focuses on identification of extracellular redox-regulated endothelial cellular function by examining mechanisms implicated in cardiovascular diseases

Patrick Tso, PhD, has contributed significantly to understanding intestinal lipid metabolism and  diet-induced obesity. Recruited to UC in 1996, Patrick Tso and colleague David Hui, PhD, have built an outstanding Lipid Research Group, recognized for its excellence by the National Institutes of Health (NIH) through awarding of a program project grant.

Together with Stephen Woods, PhD, and Randy Seeley, PhD, Dr. Tso played an important role in establishing the Cincinnati Obesity Research Center, which was initially supported by Procter & Gamble and by an  NIH program project grant. Dr. Tso was also responsible for assembling the investigative team associated with the NIH-supported Mouse Metabolic Phenotyping Center, now in its tenth year of existence.

In November 2008, Dr. Tso took on  leadership of the Physician Scientist Training Program (PSTP).  The PSTP program enjoys  success through awards  students have acquired and  awarding three additional yearly slots for two years from the NIH.

The main focus of my research effort has been to understand the metabolic basis of ischemic injury and heart failure in diabetics. In this context, our work has led to the novel finding that the glucose metabolizing enzyme, aldose reductase, assumes a critical role in the pathogenesis of myocardial ischemic injury in diabetics and non-diabetics.  Aldose reductase, both the first and rate-limiting enzyme in the polyol pathway, has been implicated in numerous diabetic complications.  Our laboratory demonstrated that substrate flux via aldose reductase adversely impacts cardiac glucose metabolism and is a mediator of ischemic injury. The work of our laboratory provided the insight into a direct link between glucose flux via aldose reductase and ion homeostasis in cardiac tissue. In subsequent studies, we provided the missing link in the ischemic heart, that is, the means by which biochemical/metabolic disturbances recruit signal transduction pathways that trigger perturbation, mitochondrial dysfunction, and cell death in the heart. 

Currently, we have focused on the ability of aldose reductase to act independent of cytosolic redox changes.  In this context, studies are in progress to link flux via aldose reductase to (a) generation of advanced glycation end product (AGE) precursors, thereby activating AGE-RAGE pathway dependent signaling for cellular injury, and (b) activating nuclear transcription factors, such as HIF-1α, that are critical determinants of cell survival. 

In our quest for exploring the impact of aldose reductase comprehensively in diabetic cardiovascular disease, we have embarked on studies investigating flux via aldose reductase in influencing atherosclerotic lesion formation and femoral artery denudation injury.  Studies are in progress to dissect the mechanisms by which aldose reductase promotes/accelerates atherogenesis. 

Driven by the complexities of mechanisms that underlie myocardial ischemic injury and diabetic cardiovascular complications, the goal of my laboratory is to elucidate the importance of aldose reductase, and AGE-RAGE in mediating ischemic injury, metabolic imbalances, heart failure and atherosclerosis.  It is my hope and expectation that interventions that block these key pathways will be employed in the near future to treat patients with evolving myocardial infarction and diabetic cardiovascular complications.
